How genetic variants affect you

Most mutations are silent. Some shape your biology in small ways. A few are decisive. Learn how penetrance and variable effects explain why the same gene can mean different things for different people.

What Is a Variant?

No two people share exactly the same DNA. The differences are tiny — your genome and a stranger's are about 99.9% identical — but that remaining 0.1% adds up to roughly 4-5 million differences, called variants.

A variant is simply a place in your genome where your DNA differs from the reference sequence. Most variants are single-letter swaps (a T where most people have a C, for example). Others involve missing letters, extra letters, or longer rearrangements.

Most Variants Do Nothing

The vast majority of variants are biologically silent. They sit in regions of the genome that don't code for anything important, or they swap one letter for another in a way that doesn't change the resulting protein. You carry millions of these, and they have no measurable effect on your health, traits, or behavior.

This is good news. It means you can't simply scan a genome and predict everything about a person — most of what's there is just background variation.

Some Variants Have Small Effects

A smaller subset of variants nudges biology in measurable ways. They might raise or lower your sensitivity to caffeine, change how efficiently you absorb a vitamin, or shift your risk for a common condition by a few percentage points.

These small-effect variants don't determine outcomes on their own — they accumulate. Your overall risk for something like type 2 diabetes is the sum of hundreds of these tiny pushes, combined with diet, exercise, and other lifestyle factors.

A Few Variants Are Decisive

A small number of rare variants, often in well-studied disease genes, can have very large effects. These are the variants that genetic counselors focus on most carefully — for example, certain BRCA1/BRCA2 changes that significantly raise breast and ovarian cancer risk.

But even decisive variants don't guarantee anything. The concept of penetrance describes how often a variant actually leads to the predicted outcome. A variant with 80% penetrance means 8 out of 10 people who carry it will show the trait — but 2 out of 10 won't.
